SQL Server does not exist or access denied Error

We are getting the following error at least twice a day now.
When this happens the web site will be down for 2 minutes and then it comes
up by itself. I though it is a timeout problem. So I changed all the timeout
values to 300 on the web server. But the problem is still there.
Any idea? Please help.
