I have a .htaccess file that works on a hosted server but not locally to
deny access to a folder. It's content simply are:

Order Deny,Allow
Deny from all

When the browsers goes to that directoryt, it lists all the files. You
can't see the .htaccess file but it is not denying access either.

Is there a setting I need to edit in the .conf file to allow this to work?

Yes. AllowOverride. See the FAQ in the Apache web site.
