I want to migrate my application from Websphere portal server 5.0 to Websphere Portal server 5.1.Is it a requirement the portlet for Websphere Portal server 5.1 should be JSR 168 compliant

You should have a look at the InfoCenter. There is a chapter that
describes how to migrate portlets. The chapter is called "Migrating from
the IBM Portlet API"
