Is this possible using WMI or any scripting mechanism?  The parties and
roles don't seem to be exposed through WMI

Yeah,
You can use the Binding file with BTSDeploy utility.
For creating Binding file with Parties use BizTalk Deployment Wizard, inside
use "Export items not assotiated with any assembly" option.
