Is there any way we can block a group of domain users from
acessgin a web site ? I tried the Grant & deny access
option for blocking domain group but it doen not work ...

Change the NTFS permissions? Either explicitly grant those users you wish to
have access Read permissions -or- if you want to allow everyone except a
particular group, put a Deny on the files for that group.
