This never happened to me with native adapters, but it sometimes happen with
custom adapters...did you repair BizTalk lately?
anyway, most chances are that all you need to do is restart the BizTalk
service(s).

if this doesn't help repair the BizTalk installation to re-register the file
adapter.

Is it a mapped drive?  BizTalk will only support full UNC paths
