Here is how I develop my BTS2004 solutions.
Development Workstation:
WinXP SP2, VS.NET 2003, SQL 2000 SP3A, BTS2004 Developer Edition.
Test Server(s):
BizTalk Machine - Win2003, BTS2004 Std. Edition
SQL Machine - Win2003, SQL 2000 SP3A
Production Servers same as Test Servers.
I develop my BTS2004 solutions in VS.NET on my development workstation,
running against a local copy of SQL 2000. I test, test, test on development
workstation and use XCOPY / BizTalk Deployment Wizard to deploy to Test
Servers. I test, test, test and then use same XCOPY / BizTalk Deployment
Wizard to deploy to production servers.
My blog has a few pointers for simple deployment and the online help has
several different deployment scenarios and tools to use depending upon your
situation.

Integration with AD is done by using domain accounts / groups during
installation. Integration with MSMQ is provided by BizTalk's new MSMQT
functionality which is explained in the online help.
DON'T GIVE UP! BTS2004 is a huge new product with many different ways to
accomplish the same thing. Finding the best way for your situation will take
some time and effort. Start with the new Microsoft Business Process and
Integration Development Center (http://msdn.microsoft.com/bpi/). Read all
the BTS related blogs possible (especially Scott Woodgate's blog at
http://blogs.msdn.com/scottwoo/) and post your questions to the newsgroups.
